Abstract [en]
Photosystem I (PSI) catalyses the transport of electrons through the membrane between plastocyanin on the luminal side and ferrodoxin on the stroma side. Plastocyanin is the electron donor and ferrodoxin is the electron acceptor. PsaF is a transmembrane protein involved in plastocyanin’s docking to PSI. At Gothenburg University there is ongoing studies of plastocyanin and the interaction between plastocyanin and PSI. Up until now they have studied isolated plastocyanin and the electron transfer from plastocyanin to PSI. The PSI complex is too big for NMR and therefore it is interesting to isolate the luminal domain of PsaF in order to perform NMR. To achieve this we constructed clones expressing a shortened version of PsaF only containing the luminal domain of PsaF; this was done through mutagenesis.
