Abstract [en]

A simple method of single-chromatography PhotosystemI (PSI)-separation without the use of ultracentrifugation was evaluated. The thylakoids were treated with the detergent n-decyl-β-D-maltopyranoside (DM), centrifuged and run in a DEAE Fast Flow column. The fractions were tested for PSI/PSII content by measuring PSI/PSII activity using an oxygen electrode and by using gel-electrophoresis. The obtained fractions seem to contain more PSI than PSII. However, this simple method does not seem to give as good separation as methods using double-chromatography.
